This plant has diverse and potential medicinal uses in traditional herbal medicine for treating skin rashes, insects and snake bites, lesions caused by herpes simplex virus, diabetes, and gout in South Asia. This study was conducted to evaluate the antioxidant, α-glucosidase and xanthine oxidase inhibitory activity of extract and fractions from Clinacanthus nutans along with phytochemical profiling by gas chromatography time of flight mass spectroscopy. Liquid-liquid partition chromatography was used to separate methanolic extract to get hexane, ethyl acetate, butanol and water fractions. The total antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazy (DPPH) radical scavenging. The antidiabetic and anti-gout activity were examined by α-glucosidase and xanthine oxidase inhibitory bioassay. The comprehensive chemical profiling of extract and fractions was carried out by gas chromatography coupled with quadrupole time-of-flight mass spectrometry (GC Q-TOF-MS). The total extractive value for methanol extraction was 12.63% (w/w) and highest fractionated value found for water fractions 31.51% (w/w) as compared to the other fractions. Moderate DPPH free radical scavenging activity was found for methanolic extract (63.07%) and among all the fractions ethyl acetate (79.98%) was the most potent fraction showed this activity. In vitro antidiabetic study of methanolic extract revealed 58.23% inhibition of α-glucosidase enzyme whereas ethyl acetate fraction produced highest inhibition of 70.77% . Poor anti-gout activity was found for methanolic extract and all the fractions except ethyl acetate fraction which showed 75.14% inhibition of xanthine oxidase enzyme.
